摘要
目的探讨白细胞介素(IL)-8和基质金属蛋白酶(MMP)-7基因多态性与早期宫颈癌的发病风险之间的相关性。方法提取早期宫颈癌组和健康对照组的外周血DNA。分别采用四引物突变扩增体系法-聚合酶链反应(ARMS-PCR)和限制性片段长度多态性法(RFLP)-PCR分析IL-8和MMP-7的基因多态性。统计学分析采用χ2检验。结果 IL-8-251AT、TT和MMP-7-181GG基因型分布频率在早期宫颈癌组中高于健康对照组(P<0.05),二者是早期宫颈癌发病的危险因素,携带有这两种基因型的患者其早期宫颈癌发病的风险可明显升高。且IL-8和MMP-7基因突变与早期宫颈癌的分期及肿瘤分化程度之间也存在显著相关性。结论 IL-8-251A/T和MMP-7-181A/G基因多态性与早期宫颈癌的发病密切相关。
Objective To determine the correlation between polymorphism at IL-8 and MMP-7 genes and risk of early stage cervical cancer. Methods Peripheral blood DNA was extracted from patients with early stage cervical cancers and controls. The incidence of IL-8 and MMP-7 gene mutations was assessed using tetra-primer amplification refractory mutation system polymerase chain reaction (ARMS-PCR) and restriction fragment length polymorphism (RFLP). The statistical analysis of the results was performed using χ2 test. Results There was a significant difference in the genotype frequencies of IL-8-251A/T and MMP-7-181A/G between the cancer group and control group (P〈0.05). Individuals with the two polymorphisms had higher risk to develop early stage cervical cancer. Moreover, IL-8 and MMP-7 polymorphisms were related to the stage of cervical cancer and the differentiation of cancer. Conclusion There was a close correlation between polymorphism at IL-8-251A/T and MMP-7-181A/G and high risk of early stage cervical cancer.
