丙型肝炎病毒核心蛋白基因序列变异分析

SEQUENCE VARIABILITY ANALYSIS OF THE HCV CORE PROTEIN GENE

为了研究西安地区丙型肝炎病毒（ＨＣＶ）基因组中Ｃ区基因的变异性，采用单链构象多态性（Ｓｉｎｇｌｅ－ＳｔｒａｎｄＣｏｎｆｏｒｍａｔｉｏｎＰｏｌｒｍｏｒｐｈｉｓｍ，ＳＳＣＰ）分析万法，对３６份ＨＣＶＲＮＡ阳性血清的ＰＣＲ产物进行分析，结果有３１份样品的ＳＳＣＰ图谱相同，对其中二份样品进行测序，证明序列是相同的；另外５价样品ＳＳＣＰ图谱不相同，其序列与前者也有差异。说明本地区ＨＣＶ基因组中Ｃ区基因存在一个主要的基因类型。该实验为本地区ＨＣＶ诊断试剂和疫苗的研究确定了研究材料。

in order to research the variability ofHCV core protein gene (HCV C gene) in Xi'an, weanalyzed 36 pieces of serum's PCR products of HCVgene by SSCP. The results showed that 31 sampleshad the same SSCP graph. We sequenced 2 PCRproducts of the 31 and found out that their se-quences were the same. The other 5 samples' SSCPgraphs were different from the 31 samples. We sequenced one PCR product of the 5 and the resultwas different from the 2 samples' we had sequenced. It proves that there is a main type of HCVC gene in Xi'an. The experiment points out a way toresearch the diagnosis kit and vaccine of HCV inXi'an.
