摘要
本文建立了高效液相色谱柱前衍生化法测定地沟油中5种生物胺含量的方法。样品经0.5mol/L盐酸提取后用丹磺酰氯衍生,采用ZORBAX 80A Extend-C1(84.6×150mm5μm),以0.1mol/L乙酸铵和乙腈为流动相进行梯度洗脱,流速1.0mL/min,柱温30℃,检测波长260nm。该方法检测限为腐胺1.2μg/g,尸胺1.4μg/g,组胺1.3μg/g,酪胺、精胺1.5μg/g。样品添加浓度为10μg/g、20μg/g时,回收率为82%～89%,相对标准偏差为1.8%～3.5%。本法为地沟油的质量鉴定提供了简便、灵敏、可靠的检测方法。
A method was established for the determination of five biogenic amines in trashy oil by high performance liquid chromatography. The samples were extracted with 0.5mol/L hydrochloric acid solution and derivatized with dansyl chloride,Using ZORBAX 80A Extend-C18 （4.6×150mm 5μm）,A gradient elution program with a mixture of 0.1 mol/L ammonium acetate and acetonitrile was used,column temperature 30℃,detection wavelength 260nm.The detection limits were 1.2μg/g for Putrescine,1.4μg/g for Cadaverine,1.3μg/g for Histamine,1.5μg/g for Tyramine and spermine.The assay was validated by analyzing samples spiked with 10,20 μg/g biogenic amines,The average recoveries and the relative standard deviations were in the range of 82%-89% and 1.8%-3.5%respectively. The article offerd a detection method for identify. This method was convenient,sensitive and reliable.
