HAPC易感性与HLA-Ⅱ类基因相关性探讨

ASSOCIATION STUDY BETWEEN HAPC SUSCEPTIBILITY AND HLA-Ⅱ GENE POLYMORPHISM

摘要 目的探讨移居高原汉族男性人群人类白细胞抗原(HLA)Ⅱ类基因中的DQA1、DQB1多态性与其发生高原红细胞增多症(HAPC)的相关性。方法以48例移居高海拔地区男性HAPC患者和48例移居同地区健康男性为研究对象,应用聚合酶链反应-序列特异性引物(PCR-SSP)技术进行HLA-DQA1、-DQB1基因分型,使用SPSS软件分析HLA-DQA1、-DQB1多态性的分布频率及其组间差异。结果移居高海拔地区的HAPC患者组的HLA-DQA1*0601分布频率与同地区健康对照组相比有统计学意义(χ2=5.547,P<0.05)。而HAPC组和对照组在DQB1*0303、DQB1*0401、DQB1*0501等位基因上未显示显著性差异。结论 HLA-DQA1*0601位点可能与HAPC的易感性关联。 Objective To explore whether HLA-DQA1,-DQB1 polymorphisms of Human Leukocyte Antigen (HLA) Class Ⅱgene are associated with the susceptibility of High Altitude Polycythemia (HAPC) in Chinese Han nationality immigrated to plateau area. Methods Peripheral blood samples from 48 cases of HAPC male patients and 48 healthy male individuals from same area with HAPC group were used to genotype HLA-DQA1,-DQB1 genes with polymerase chain reaction using sequence specific primer method (PCR-SSP). The frequency distributions of genotypes between two groups were analyzed by SPSS software. Results The distribution frequency of HLA-DQA1*0601 alleles in HAPC patients group were significantly different from that in control group,Its χ^2=5.547,P=0.019. But there were no frequency differences in DQB1*0303、 DQB1*0401、 DQB1*0501 alleles. ConclWusion HLA-DQA1*0601 alleles may associated with the susceptibility of HAPC.
