地骨皮提取液对H_2O_2诱导的人脐静脉内皮细胞凋亡的影响 被引量:7

摘要 目的:探讨地骨皮醇提液对H2O2诱导的HUVEC细胞凋亡的影响,探讨地骨皮醇提液对血管内皮的保护作用,为研究糖尿病并发症治疗的新药提供新的理论基础。方法:将HUVEC细胞分为正常对照组、高药物浓度组、H2O2组和药物预处理+H2O2作用组,作用18-24h后,用显微镜观察细胞形态改变,用流式细胞仪通过Annexin V/PI双染法检测各组细胞的凋亡百分比,用RT-PCR检测Bcl-2基因的表达变化。结果:地骨皮醇提液有效降低H2O2引起的细胞凋亡率;地骨皮醇提液可以增加凋亡相关基因Bcl-2的表达。结论:地骨皮醇提液对H2O2诱导的HUVEC细胞凋亡有不同程度的抑制作用,对血管内皮具有保护作用。 Objective:To explore the protection effects of Chinese herbs Cortex lycii on H2O2-induced apoptosis in the Human Umbilical Vein Cells.Methods:Make four groups of the HUVEC as normal cells group,H2O2-only deal cells group and Cortex lycii+H2O2 acting cells group.After 18~24h,flow cytometry by Annexin V/PI double stain was performed to quantify the apoptotic cells.The expressions of Bcl-2 gene was tested by RT-PCR.Results:The HUVEC was treated with Cortex lycii,the apoptosis rate decreased in different degrees.RT-PCR detection showed that H2O2 inhibit the expression of the Bcl-2 gene,and when they were treated by different concentrations of Cortex lycii,the expression of the gene increased following the different concentrations of the herbs.Conclusion:Cortex lycii could prevent H2O2-induced HUVEC cells from apoptosis in different degrees,of which the activation of the expression of Bcl-2 gene.
