摘要
[目的]介绍由孤雌生殖诱导系诱导的玉米单倍体的鉴定方法。[方法]利用孤雌生殖诱导系诱导的单倍体为材料,根据籽粒Navajo标记和ABP1紫色植株显性双标记系统对诱导产生的单倍体进行鉴定,对根尖细胞染色体数目进行显微观察。[结果]籽粒Nav-ajo标记和ABP1紫色植株标记可淘汰大量杂交二倍体,细胞学鉴定可直接验证其他表型鉴别方法的准确性。[结论]以上方法对于孤雌生殖单倍体的鉴别是可靠的。这些单倍体鉴别方法各有优缺点,综合利用它们可以提高单倍体鉴定效率。
[Objective] The aim was to set up an identification method for maize monoploid induced by parthenogenesis inducer.[Method] By using monoploid induced by parthenogenesis inducer as material,the induced monoploid was identified on the basis of grain-Navajo marker and ABP1 purple plant-dominant double markers system and the chromosome number in root-tip cells was observed through microscope.[Result] A lot of hybrid diploids could be eliminated by using grain-Navajo marker and ABP1 purple plant-markers.The accuracies of the other phenotype-identification methods could be verificated directly through cytological identification.[Conclusion] The identifications by using the above methods on parthenogenesis monoploid were reliable.These monoploid-identification methods had their peculiar advantages and disadvantages and the monoploid-identification efficiency could be enhanced by utilizing them comprehensively.
