摘要 [目的]建立土人参愈伤组织的诱导、增殖继代培养及其细胞悬浮培养的技术体系。[方法]研究不同植物激素组合对叶片、茎段愈伤组织诱导的影响;采用液体振荡培养法测定叶片愈伤组织细胞悬浮生长曲线。[结果]叶片最易诱导产生愈伤组织,最佳诱导、增殖培养基为MS+2,4-D1.0 mg/L+NAA0.5 mg/L+6-BA0.5 mg/L(pH5.8)。由叶片愈伤组织建立的细胞系生长呈"S"型曲线,最适继代周期为15 d。[结论]以叶片为外植体经愈伤诱导、增殖培养可建立良好的细胞悬浮系。 [Objective] The aim was to establish the technical systems of callus induction,multiplication subculture and the cell suspension culture of T.paniculatum.[Method] Effects of different plant growth regulators on callus induction from leaf and stem segment were studied The cell suspension culture was established from leaf calluses and the growth curve was tested by liquid shake culture methed.[Result] Leaf was the optimal explant for callus induction.The optimal medium of multiplication subculture for leaf calluses was 2,4-D 1.0 mg/L + NAA 0.5 mg/L+6-BA 0.5 mg/L(pH 5.8).The cell growth of T.paniculatum with "S" type curve was observed and optimal subculture cycle of suspension cell was 15 days.[Conclusion] The callus induced from leaf explant was excellent for establishing cell suspension of T.paniculatum.
