摘要 [目的]探讨碱提香菇菌丝体粗多糖抗氧化能力的影响因素。[方法]按Sevag法去除粗多糖中蛋白质,以考马斯亮蓝法、苯酚-硫酸法和DPPH法分别测定碱提香菇菌丝体粗多糖在脱蛋白过程中多糖含量、蛋白含量和清除自由基能力的变化。[结果]碱提香菇菌丝体水溶性多糖经Sevag洗脱后,多糖含量降低了33.10%;蛋白含量降低了97.08%;自由基清除率降低了74.38%。[结论]碱提香菇菌丝体多糖具有较高的抗氧化能力,并与其糖蛋白浓度成正相关,过分脱除香菇菌丝体多糖提取物结合态蛋白会降低其抗氧化活性。 [Objective] The research aimed to discuss the influencing factors of the antioxidative capacity of crude polysaccharides extracted from Lentius edodes mycelia with alkali.[Method] Sevag method was adopted to remove protein.Coomassie brilliant blue method,phenol-sulfuric acid method and DPPH method were used to determine the changes of polysaccharide content,protein content and scavenging ability of free radicels in the process of deproteinization.[Result] After water-soluble polysaccharide extracted from Lentius edodes mycelia with alkali was eluted by Sevag method.The content of polysaccharide reduced 33.10% and protein content reduced 97.08%.The scavenging rate of free radicels reduced 74.38%.[Conclusion] The polysaccharides extracted from Lentius edodes mycelia with alkali had higher antioxidation capacity.And it had positive correlation with the glycoprotein concentration.The antioxidant activities would be reduced after excessive deproteinization of the binding protein of polysaccharides extracts from Lentius edodes mycelia.
