摘要
[目的]优化大批量快速提取白僵菌DNA的方法。[方法]对DNA沸水抽提方法进行改进,通过PCR扩增仪对DNA提取液进行加热,迅速完成DNA的提取过程。[结果]通过大批量快速提取真菌基因组DNA方法提取的DNA质量可满足RAPD扩增分析的需要。22个试验菌株均扩增出清晰的条带,清晰条带数量为2~6条不等,条带大小主要集中在450~800 bp;该方法提取的DNA也完全可以满足SCAR扩增的需要,扩增出的用于标记F263菌株的特异性DNA条带十分清晰。[结论]该研究为大批量快速提取真菌基因组DNA提供了较为理想的方法。
[Objective]The aim was to optimize the bulk and rapid extraction method of DNA from Beauveria bassiana.[Method]For improving boiling-water extraction of DNA,the DNA extract was heating by using PCR instrument and the extraction process was finished rapidly.[Result]The quality of DNA obtained through bulk and rapid extraction of genomic DNA from fungi could satisfy the demand of RAPD amplification and analysis.The clear bands were amplified from 22 tested strains,and the band quantities were different in 2-6 and the band sizes were mainly concentrated in 450-800 bp.The DNA extracted by using this method also could satisfy the demand of SCAR amplification completely.The amplified specific DNA bands,which were used to mark the strain F263,were very clear.[Conclusion]This research supplied relatively perfect method for bulk and rapid extraction of genomic DNA from fungi.
