摘要
目的研究二乙酰吗啡致大鼠心律失常类型及膜电位变化对心律失常的影响。方法采用二乙酰吗啡剂量递增法,实验组大鼠为Wistar大鼠,二乙酰吗啡使用量从5 mg/kg开始,每日3次,以后每日递增2.5 mg/kg,观察盐酸纳洛酮催瘾后的戒断症状,判断二乙酰吗啡致大鼠成瘾是否成功建立。实验组大鼠成瘾后继续增加二乙酰吗啡使用量,进一步建立实验组大鼠的心律失常模型,直至实验组大鼠死亡,分析大鼠死亡前心律失常的类型;对心律失常大鼠采用膜片钳技术检测膜电位变化。结果二乙酰吗啡致大鼠出现各种心电图变化及各种心律失常类型,心肌细胞动作电位时程中APD90显著缩短(P<0.05)。结论二乙酰吗啡致大鼠心肌细胞膜电位的改变为心律失常的发生原因之一,成瘾后的大鼠死亡的心律失常类型为临床治疗及法医学检案提供理论依据。
Objective To study proarrhythmia types and the effects of membrane potential proarrhythmia by diacetyl of morphine in rats.Methods The steadily increase way about diacetyl of morphine dosage was used.The Wistar rats were divided into two groups,the experiment and the control.The first dosage was 5 mg/kg,three times everyday,increased 2.5 mg/kg everyday,observed the symptom of withdraw after injected naloxin in order to judge whether the model success.When the experiment rats were abused,we continud to add the diacetyl of morphine dosage in order to establish the experiment rats proarrhythmia model,when the rats dead,we analysised the rats proarrhythmia types.When the proarrhythmia appeared,we detected the change of the cell membrane potential by the technology of patch clamp.Results When the experiment rats were abused by diacetyl of morphine,we found a series of animal behaviors,and when the rats proarrhythmia appeared we found all kinds of proarrhythmia about rats death.They were significantly different(P〈0.05),the APD90 curtailed evidently in the action potentia.Conclusion The occured of proarrhythmia could be come into being by the change of membrane potential.We could receive all kinds of proarrhythmia by these abused rats,these research findings could provide basis theories for clinical treatment.
