甘薯优良品种试管苗培养系的建立 被引量:7

摘要 从田间选取甘薯优良品种12份,采用MS基本培养基进行无菌试管苗培养.结果表明,餐具洗涤剂浸泡15min,75%乙醇浸泡30s,再用含吐温20的01%氯化汞溶液浸泡10min的杀菌效果最好.采用快速转移接种除菌法可使外植体污染率大大降低,平均为5%以下.无菌试管苗离体生长特性研究表明,强光有利于试管苗的生长,不同甘薯基因型在相同培养基和相同培养条件下其生长状况存在较大差异. The explants of 12 superior sweetpotato cultivars were selected from field and cultured on MS medium to obtain sterile cuttings.The results showed that the best treatment for sterilization was that,explants were dipped in detergent for 15 minutes,then in 75% alcohol for 30 seconds,followed by 0 1% HgCl 2solution with Tween 20 for 10 minutes.Pollution rate of explants was greatly decreased to 5% by rapidly cutting the new sprouts and transfering them to fresh media.The intensive light was benefit to growth of in vitro cuttings,some obvious differences were observed in various sweetpotato genotypes,even which were cultured in the same medium and in the same culture condition.
