摘要
目的:建立人腱细胞体外培养系,为肌腱愈合研究提供一种模型。方法:用贴块法培养人胚胎腱细胞,并观察了细胞生长、生化及形态特征,用免疫组化染色鉴定细胞表型。结果:人胚腱细胞在体外呈长梭形,胞质均匀透明,汇合后呈平行致密排列并表现接触抑制特征,传代后生长特征仍保持稳定,细胞免疫组化染色Vimentin及Ⅰ型胶原呈阳性,而Fn及Ⅲ型胶原呈阴性,人胚腱细胞最适合在含10%胎牛血清及5%胎儿血清的DMEM培养基中生长。
Objective: To establish the in vitro model of tendon cell biology study. Methods: Human embryo tenocytes were cultured using the explant technique.The morphology and growth and biochemistry characteristics of primary and passaged cells were observed. The tenocytes were identified by Vimentin. Fn、 type Ⅰ、Ⅲ collagen McAb immunohistrochemistry. Results: The explant methed was optimal for tenocytes culture. The cells reached confluency usually after 6~7 days and presented parallel rank. The primary cells was stained positively for Vimentin、 type I Collagen and negatively for Fn、 type Ⅲ collagen. Add in 5% fetal human serum (FHS) to DMEM medium contained 10% Fetal bovine serum (FBS) was favourable for cells growth. Conclusions: The cultured human embryo tendon cells provide a sort of experimental materials for tendon healing and tissue-engineering study.
