摘要
用蒽酮-硫酸法和3,5-二硝基水杨酸(DNS)法测定多糖含量。以多糖得率为指标,采用正交实验法分别对水提和醇沉过程的条件进行优选,最佳水提工艺为:提取温度100℃,料液比1:10,提取2次,每次4h;最佳醇沉工艺为:浓缩液加入3倍体积100%乙醇,沉淀4h,此工艺条件下多糖得率为1.58%。该工艺简单,适于杜仲皮多糖提取。
The content of polysaccharide was determined by 3,5-dinitrosalicylic acid colour reaction and anthrone-sulfuric colorimetric method.Taking extracting yield of polysaccharide as investigation index,the technology of water extraction and ethanol precipitation was optimized by orthogonal experiment.The optimum water extraction conditions were that the material-water ratio was 1:10,the extracting temperature was 100 ℃,the extracting time was 4 h,the extracting number of times was 2 times.The optimum ethanol precipitation process was as follows:the concentrated solution was precipitated with 3 times of 100% ethanol for 4 h.The extracting yield of polysaccharide was 1.58% under the former conditions.The method was simple and suitable to extract polysaccharide from the bark of Eucommia ulmoides Oliver.
