HPLC法快速检测大豆籽粒中大豆低聚糖的含量 被引量:15

The method for rapid detection of soybean oligosaccharide content by HPLC

摘要 建立了高效液相色谱法同时快速测定大豆籽粒中大豆低聚糖的3种组分蔗糖、棉籽糖和水苏糖含量的检测方法。采用0.5量程示差折光检测器,4.6mm×250mm,5μm瑞典Kromasil-NH2原装进口柱,流速1.0mL/min,检测温度35℃,流动相乙腈:水=60:40,10μL进样量。在上述检测条件下,最低检出限蔗糖为0.06×10-6g、棉籽糖为0.07×10-6g、水苏糖为0.06×10-6g,标准曲线相关系数分别为0.9965、0.9996和0.9994,平均回收率分别为99.3%、94.7%和95.6%,RSD(n=10)分别为1.73%、2.01%和0.80%。该方法简单,快速,准确,可用于大豆籽粒中低聚糖含量的快速分析。 Established a high-performance liquid chromatography method for detecting the content of sucrose、 raffinose and stachyose in soybean grains.Use refractive,index detector with range 0.5,Kromasil-NH2 column imported from Sweden,and its model is 4.6 mm×250 mm 5 μm,the velocity of flow is 1 mL/min,the detection temperature of column is 35 ℃,and the acetonitrile:water=60:40.Under the conditions of the above-mentioned test,the minimum detection limit of source is 0.06×10-6 g,the minimum detection limit of raffinose is 0.07×10-6 g,and stachyose is 0.06×10-6 g.The correlation coefficients of standard curves were 0.9965,0.9996 and 0.9994.The average recoveries were 99.3%,94.7% and 95.6%.The RSD of samples with 10 tests are 1.73%,2.01% and 0.80%.The method is simple,rapid,accurate,and can be used in the oligosaccharide content in soybean quick analysis.
