六味地黄汤对雷公藤多甙损伤小鼠精子的影响

Influence of Liuwei Dihuang Decoction on Sperm Lesion Induced by Multiglycosides of Tripterygium Wilfordii Hook in Mice

雄性小鼠灌胃雷公藤多甙２０、３０ｍｇ／ｋｇ，１４ｄ或２１ｄ均能引起精子损伤。六味地黄汤（剂量以ｇ生药／ｋｇ表示）以１５ｇ／ｋｇ或２５ｇ／ｋｇ灌胃能对抗雷公藤多甙以２０ｍｇ／ｋｇ灌胃１４ｄ所造成的精子损伤。通过对生育力的研究发现，雷公藤多甙以２０ｍｇ／ｋｇ灌胃雄性小鼠１４ｄ不能明显减少受孕雌性小鼠的只数，但能明显减少孕鼠的胎仔数。六味地黄汤分别以１５、２５ｇ／ｋｇ灌胃均能对抗雷公藤多甙降低胎仔数的作用。拆方研究表明：六味地黄汤全方与三泻（泽泻、丹皮、茯苓）、三补（熟地、山药、山茱萸）均有明显对抗雷公藤多甙损伤精子的作用。三泻与全方作用相当，而三补作用稍弱。

Male mice ig multiglycosides of Tripterygium wilfordii Hook (MGTW) 20 mg/kg or 30 mg/kg for 14 d or 21 d could produce sperm lesion. Liuwei Dihuang Decoction (LDD) ig 15 g/kg or 25 g/kg antagonized the sperm lesion caused by ig MGTW 20 mg/kg for 14 d. Meanwhile, three tonics (Radix Rehmanniae Preparata, Fructus Corni, Rhizoma Dioscoreae) and three purgatives (Poria, Cortex Moutan Radicis, Rhizoma Alismatis) also exerted the antagonistic effect. Further, the effect of three purgatives was stronger than that of three tonics. Through the study of fertility, we found that MGTW (20 mg/kg×14 d, ig) did not decrease the number of female pregnant mice, but significantly decreased the fetal number. This effect could be antagonized by ig LDD 15 g/kg or 25 g/kg.
