奥硝唑凝胶剂体内外抗阴道毛滴虫作用 被引量:4

Effect of ornidazole gel on Trichomonas vaginalis in vivo and in vitro

摘要 目的:研究奥硝唑凝胶剂在体内外对阴道毛滴虫的作用。方法:体外实验将阴道毛滴虫培养后分别接种到含不同浓度奥硝唑凝胶、奥硝唑和奥硝唑栓剂的细胞培养板孔中,于不同时间在倒置显微镜下观察虫体形态变化,计数死虫百分率。体内实验将小鼠阴道内连续多日注入药物和虫体,停药1d后取阴道液涂片观察有无活动的阴道毛滴虫。结果:体外实验各组药物作用24h后,各种药物的最低有效抑虫浓度(虫体100%死亡)依次为:奥硝唑凝胶0.0025g.L-1,奥硝唑原料0.0049g.L-1,奥硝唑栓剂0.0195g.L-1。体内试验中连续5d注入药物和虫体的奥硝唑原料药组,奥硝唑凝胶剂组,奥硝唑栓组,镜检均为阴性(0/20)。而无药对照组100%(20/20)阳性。结论:奥硝唑凝胶有很好的体内外抗阴道滴虫作用,其作用优于奥硝唑栓。 OBJECTIVE To study effect of ornidazole gel on Trichomonas vaginalis in vivo and in vitro. METHODS In vitro experiment, Trichomonas vaginalis was cultured and inoculated into wells of microplate, which contained different concentrations of ornidazole gel, ornidazole, ornidazole suppository, respectively. The morphological changes of parasite were observed under invert microscope and the death rate was calculated after different incubation time. To monitor the effect of ornidazole on parasite in vivo,drugs(ornidazole gel,ornidazole,ornidazole suppository) and the organism were injected into vagina of mice for several days. Surviving organisms in vaginal secretions of mice were examined at the second day after last injection. RESULIS After 24 h of incubation, minimum killing concentrations(concentration of killing all organisms) of different groups in vitro were 0. 002 5 g·L ^-1 ornidazole gel,0. 004 9 g·L ^-1 ornidazole,0. (119 5 g·L ^-1 ornidazole suppository,respectively. The parasites in groups of ornidazole,ornidazole gel and ornidazole suppository were all negative (0/20) in vivo after injected with organisms and drug for 5 d, while the control group was 100% positive (20/20). CONCLUSION ornidazole gel is efficacious against Trichornonas vaginalis both in vivo and in vitro ,and it's better than ornidazole suppository.
