摘要
目的:研究洛美沙星(LFX)和环丙沙星(CPLX)对小鼠肝肾的毒性作用。方法:分别单次ipLFX和CPLX各0.375,0.75和1.5mg/10g体重,所有小鼠15d后处死。结果:部分肝脏及肾脏与周围组织粘连;CPLX(1.5mg/10g)组第3蛋白电泳带含量比对照组增加48.1%(P<0.05);肝重/10g体重及肝功能无明显变化;光镜下,除LFX(0.375mg/10g)组外,各组肝细胞有不同程度变性和坏死;LFX(1.5mg/10g)组1例肾脏为慢性间质性肾炎改变。结论:LFX和CPLX对小鼠肝脏有潜在的毒性作用,且无明显的剂量依赖性。
OBJECTIVE:To study the liver and renal toxic reactions of lomefloxacin and ciprofloxacin in mice.METHODS:A single dose of 0.375,0.75 and 1.5 mg/10 g body weight of lomefloxacin and ciprofloxacin were given by intraperitoneally injection to mice. After 15 days of administration all animals were killed by decapitation.RESULTS:The liver and kidney of some animals in treated groups were adhered to the surrounding tissues.The third serum fractions in ciprofloxacin (1.5 mg/10 g body weight)treated mice increased 48.1%( P <0.05)in comparison with that of saline treated mice.No significant changes were observed in the ratio of liver/10 g body weight and liver function.Hepatocyte degeneration and necrosis of various degrees could be seen by light microscopy in lomefloxacin treated group. One case of chronic interstitial nephritis was observed in lomefloxacin treated group.CONCLUSION:Lomefloxacin and ciprofloxacin have potential toxicities to liver and kidney in mice but it seems unlikely to be dose dependence.Therefore,these drugs should be used with caution in patients with liver or renal dysfunction.
