摘要
利用海南橡胶树多主棒孢培养液,通过透析、冷冻干燥浓缩、硫酸铵沉淀以及离子交换层析等方法对粗毒素进行了浓缩与提纯,获得的粗毒素通过生物测定和聚丙烯酰胺凝胶电泳验证。结果显示,该方法能获得较纯的具有活性的蛋白毒素,其在凝胶电泳中显示分子量约为14.4ku。
A toxic substance was produced by Corynespora cassiicola, one of the main pathogens affecting rubber tree (Hevea brasiliensis). Crude toxin was extracted from culture filtrate of HC-10 strain in the study. The toxin extract protocol involved dialysis, freeze-dried, ammonium sulfate precipitation, ion-exchange and gel filtration chromatography. Extracted toxin was detected by leaf puncture bioassay and polyacrylamide gel electrophoresis, and the results showed the molecular weight of the toxin protein was about 14.4 ku.
