摘要
目的:从细胞凋亡角度探讨Sirt1在银屑病发病中的作用。方法:采用密度梯度离心法分离30例银属病患者和10例健康对照者外周血单个核细胞,实时定量RT-PCR(real-time RT-PCR)法检测PBMC细胞组蛋白去乙酰化酶Sirtl mRNA表达水平。结果:银屑病患者PBMC细胞Sirtl mRNA表达水平较正常对照组明显增加。结论:推测银屑病的角质形成细胞增殖可能与凋亡抑制因子Sirtl关系密切。
Objective:To investigated the role of Sirtl in the pathogenesis of psoriasis. Methods:30 psoriasis patients and 10 healthy controls were recruited. PBMC were isolated by Ficoll-Hypaque density gradient centrifugation, the mRNA levels of Sirtl genes were measured by real-time quantitative polymerase chain reaction. Results:Sirt1 mRNA levels were significantly increased in psoriasis PBMC compared with healthy controls (P〈0.001). Conclusion:Sirt1 can promote the proliferation of keratinocytes and maintain the hyperproliferative state of psoriatic epidermis,which may partly by apoptosis interference.
