摘要
目的:高速逆流色谱分离得到高纯度的地龙中抗血栓的活性成分。方法:利用高速逆流色谱,以乙酸乙酯-甲醇-水(体积比为3:3:3)为两相溶剂系统,在主机转速1 000r/min、流动相流速2.0ml/min、检测波长254nm条件下进行分离制备,6h内从1kg地龙粗提物中一步分离制备得到商纯度的地龙中抗血栓的活性成分。结果:分离物纯度均达98.1%以上,各化合物结构经质谱鉴定。结论:利用高速分离可以得到高纯度的地龙中抗血栓的活性成分进行快速的分离和纯化。
Objective:High purity the active ingredient of anti-thrombosis in lumbricus by using high speed eountereurrent chromatography.Method:In high-speed countercurrent chromatography,ethyl acetate-methanol-water(volume ratio 3:3:3)for the two-phase solvent system,the host speed 1000 r/min, flow rate of 2.0ml/min,detection wavelength 254 nm conditions separation under preparation,6h from lkg to within the crude extract of step Lung Isolation and Preparation of high purity obtained Dragon of the active ingredient in anti- thrombosis.Results:The isolates reached 98.1% purity or more,the compound structUre by mass spectrometry.Conclusion:High-speed separation can be long in the land of high pUrity antithrombotie active ingredients for rapid separation and purification.
