Influence of PI3K on the migration of endothelial cells mediated by IL-8/Rac1 signaling pathway

摘要 目的研究磷脂酰肌醇-3激酶(PI3K)对IL-8/Rac1信号通路所介导的内皮细胞迁移的影响。方法选择P13K抑制剂wortmannin及划痕创伤愈合修复分析,研究不同wortmannin的作用浓度和不同时间对IL-8/Rac1信号通路诱导的内皮细胞迁移的影响。结果不同wortmannin的作用浓度和不同时间预处理均可抑制IL-8/Rac1信号通路介导的内皮细胞迁移,其中wortmannin预处理浓度为100nmol/L,预处理时间为20min时IL-8诱导的内皮细胞迁移水平最低。结论P13K能够影响IL-8/Rac1信号通路介导的内皮细胞迁移;P13K抑制剂wortmannin的最佳作用浓度为100nmol/L,最佳作用时间为20min。 Objective To study the influence of phosphatidylinositol 3-kinases (PI3K) on the migration of endothelial cells mediated by IL-8/Racl signaling pathway. Methods Different concentration and duration of PI3K inhibitor, wortmannin,was used to inhibit PI3K signaling pathway. Wound healing assay was performed to evaluate the effect of PI3K on the migration of endothelial cells mediated by IL-8/Rac 1 signaling. Results The migration of endothelial cells was inhibited significantly by different concentration and different time of PI3K inhibitor wortmannin. The results of wound repair area and cell migration distance showed that 100 nM wortmannin and 20 min duration exhibit the best inhibitory effects on cell migration. Conclusion The migration of endothelial cells mediated by IL-8/Racl signaling pathway could be influenced by PI3K. 100 nM wortmannin and 20 min duration showed the maximal inhibitory effects on endothelial cell migration.
