Effect of male contraceptive AF-2364 on germ cells in male rabbits

目的:观察男性避孕药AF-2364对雄兔生殖细胞的组织学和超微结构的影响。方法:将27只雄兔随机分为对照组、静脉给药组和灌胃给药组,静脉给药组和灌胃给药组分别给予AF-2364 25 mg/kg每周1次,连续4周;每隔2周,各组随机处死3只兔子,取睾丸、附睾等组织;睾丸电镜标本用2.5%戊二醛固定,制作成组织标本,用H-7500透射电镜观察;光镜标本用Bouin液固定,用OLYMPUS CX31电子显微镜观察。结果:光镜检查显示给药后生精细胞脱落,生精上皮变薄,附睾管内精子减少。电镜检查显示精子核头部溶解,尾部中段线粒体数目减少,生精细胞之间以及与支持细胞间的连接点破坏,间隙增大。结论:AF-2364能有效、可逆地抑制兔睾丸精子的发生。

Objective：To observe the effect,hitological and ultra-structrue changes of AF-2364 on germ cells in male rabbit.Methods：Twenty seven male rabbits were separated into control group,intravenous administration group and intragastric administration group at random,for intravenous administration group or intragastric administration group,each rabbit was administered AF-2364（25 mg/kg,once per week） for four weeks;3 rabbits were killed every two weeks in three groups at random,took out the testis and epididymis;testis for electron microscope observation fixed in 2.5%glutaric dialdehyde,the specimens of electron microscope observed with H-7500 transmission electron microscope;the testis fixed in Bouin,the specimens of light microscope observed with OLYMPUS CX31 microscope.Results：Optical microscope test showed rapid exfoliation of spermatogenic cell,spermatogenic epithelium became thin,sperm reduced in epididymal duct.Electron microscope test showed sperm nucleus dissolved,mitochondrion decreased.junction point of spermatogenic cell and sertoli cell destroyed the gap scale-up.Conclution：AF-2364 induced inhibition of spermatogenesis is effective and reversible.
