微波辅助萃取-高效液相色谱法测定金银花药材中槲皮素和木樨草素的含量 被引量:8

Study on microwave assisted extraction HPLC method for determination of two flavonoids compunds of Flos lonicerae

摘要 目的建立微波辅助萃取-高效液相色谱(MAE-HPIE)法分析金银花药材中槲皮素和木樨草素的含量。方法采用Diamomil C18柱(150mm×4.6mm,5μm),以甲醇-0.2%三氟乙酸(55:45,V/V)为流动相,流速1.0mL·min^-1,检测波长350nm。微波辅助萃取条件:2.5mol·L^-1盐酸甲醇溶液10mL,微波功率为360W,提取时间为3min。结果槲皮素在5-100mg·L^-1内线性关系良好(r=0.9995),平均加样回收率为92.35%,RSD为1.33%(n=3);样品的日内和日间RSD分别为0.44%和1.61%。木樨草素在0.25~5mg·L^-1内线性关系良好(r=0.9999),平均加样回收率为92.40%,RSD为1.13%;日内和日间RSD为0.79%和2.60%。结论该方法快速、准确,操作便捷,适用于提取分析和测定金银花中槲皮素和木樨草素的含量。 AIM To develop a microwave assisted extraetion-HPLC method for the determination of two flavonoids eompunds of Flos lonicerae. METHODS Quereetin and luteolin in Flos lonicerae were determined by HPLC on Diamonsil C18 column ( 150 mm × 4.6 mm,5 μm) with methanol - 0.2% trifluoroaeetie acid (55:45, V/V)as the mobile phase. The detection wavelength was 350 nm. The flow rate was 1.0 mL· min^-1. Quereetin and luteolin were both extracted by microwave assisted extraction with 10 mL 2.5 mol·L^-1 methanol-ehlorhydrie acid using condition of 360 W and 3 rain. RESULTS The linear range of quereetin was 0.25 - 5 mg·L^-1 ( r = 0.999 9) and the linear range of luteolin was 5 - 100 mg·L^-1 ( r = 0. 999 5). The average recovery and RSD value of quereetin were 92.35% and 1.33% ( n = 3) ; the average recovery and RSD value of luteolin were 92.40% and 1.13% ( n = 3), respectively. The RSD of intra-day and inter-day of quereetin were 0.44% and 1.61%, respectively; the RSD of intra-day and inter-day of luteolin were 0. 79% and 2.60%, respectively. CONCLUSION The method is simple, precise and accurate, and ean be used to determine and analyze quereetin and luteolin in Flos lonicerae.
