趋化因子及其受体CXCL16/CXCR6体外促前列腺癌增殖、侵袭作用 被引量:2

Effects of CXCL16/CXCR6 Axis on Proliferation and Invasion of Human Prostate Cancer Cell Line in Vitro

摘要 目的:探讨趋化因子及其受体CXCL16/CXCR6在人前列腺癌细胞系体外增殖侵袭中的作用。方法:免疫细胞化学技术检测CXCL16/CXCR6在人前列腺癌细胞系DU145中的表达;MTT法分析CXCL16/CXCR6对DU145细胞的促增殖作用;迁移、侵袭实验分析CXCL16对DU145细胞体外侵袭能力的调节作用。结果:DU145既表达CX-CR6蛋白,也表达CXCL16蛋白;外源性CXCL16可明显提高DU145细胞体外增殖活力和促进DU145细胞的体外迁移、侵袭能力,CXCL16中和抗体可以有效消除CXCL16对细胞的促侵袭作用,但CXCL12或CXCR4中和抗体不能阻断CXCL16的促侵袭作用。结论:CXCL16/CXCR6可能是参与前列腺癌转移的重要趋化因子配受体对。 Objective: To explore the effects of CXCL16/CXCR6 axis on the proliferation and invasion of human prostate cancer cell line DU145 in vitro.Methods: The expression of CXCL16/CXCR6 in DU145 cells was determined by immunocytochemistry.The MTT assay was used to assess the effect of CXCL16/CXCR6 on proliferation of DU145 cells.And then the effects of CXCL16 on the migration and invasion of human DU145 cells were investigated by Matrigel invasion assay.Results: The expressions of both CXCL16 and CXCR6 proteins were detected in DU145 cells.The human recombinant CXCL16 protein could promote the proliferative ability of DU145 cells.In addition,CXCL16 could promote the migration and invasion of DU145 cell lines in vitro,in which the effect of promoting by CXCL16 could not be influenced by CXCL12 or CXCR4 neutralizing antibody.Conclusion: CXCL16/CXCR6 may be an important chemokine/chemokine receptor pair which independently play a significant role in the metastasis of prostate cancer.
