摘要
目的观察大黄酚对β-淀粉样肽(Aβ25-35)致AD小鼠学习记忆障碍及脑组织过氧化氢(H2O2)含量、过氧化氢酶(CAT)、Na+-K+-ATP酶和Ca2+-ATP酶活性的影响。方法给小鼠一次性脑室内注射凝聚态Aβ25-353μg(1.0mmol/L)造AD模型,造模后腹腔注射药物或溶剂,连续注射14d。采用Y-迷宫法测试小鼠学习记忆能力,每天每鼠训练10次,连续测试5d,以正确反应次数作为观测指标。用比色法测定小鼠脑组织H2O2含量、CAT、Na+-K+-ATP酶和Ca2+-ATP酶活性。结果与对照组相比,模型组正确反应次数降低,脑内H2O2含量升高;CAT、Na+-K+-ATP酶和Ca2+-ATP酶活性显著降低;与模型组比较,大黄酚各组小鼠正确反应次数从测试2d起明显增加,小鼠脑内H2O2含量明显降低,而CAT、Na+-K+-ATP酶和Ca2+-ATP酶活性显著升高。结论大黄酚对Aβ25-35致AD小鼠记忆障碍有保护作用,其作用机制可能与抗氧化、改善能量代谢有关。
Objective To observe the effects of chrysophanol on abilities of learning and memory,contents of H2O2,activities of catalase (CAT),Na+-K+-ATPase and Ca2+-ATPase in brain tissue in AD model mice induced by Aβ25-35. Methods The aggregated Aβ25-35 3 μl (1 mmol/L) was intracerebroventricularly injected once into mice to make AD model. The solvent was administered by ip for control and model group,while chrysophanol (10.0,1.0,0.1 mg/kg) was injected for chrysophanol groups for 14 days. The abilities of learning and memory in mice were observed through Y-maze test for 5 days,in which every mouse was disciplined 10 times every day and the number of correct response was as observed indicators. Then the contents of H2O2 and the activities of CAT,Na+-K+-ATPase and Ca2+-ATPase in brain tissue in mice were detected through colorimetry. Results Compared with control group,the number of correct response was decreased,the contents of H2O2 were increased,and activities of CAT,Na+-K+-ATPase and Ca2+-ATPase were decreased significantly in model group. While the number was higher in chrysophanol group (10.0,1.0,0.1 mg/kg) from the second day,the contents of H2O2 were lower,and activities of CAT,Na+-K+-ATPase and Ca2+-ATPase were higher than in model group significantly. Conclusions Chrysophanol shows protective effects on learning and memory impairment in AD mice induced by Aβ25-35. Possible mechanisms are involved in the actions of antioxidation and improving energy metabolism.
