针灸预处理对Alzheimer大鼠Wnt_1表达的影响 被引量:17

摘要 目的建立老年性痴呆(AD)大鼠模型,探讨针灸预处理对AD模型大鼠Wnt1信号转导通路的影响及防治AD的机制。方法建立AD大鼠模型,随机分为正常组、假手术组、模型组、预艾灸组、预电针组、预电针加灸组。通过实时定量PCR技术检测Wnt1基因表达。结果模型组大鼠海马Wnt1信号异常减弱,与正常组和假手术组比较差异显著(P<0.01)。而预艾灸组、预电针组、预电针加灸组大鼠海马Wnt1的信号均有不同程度的增强。结论针灸防治AD的作用机制可能是通过调节Wnt信号转导通路影响学习记忆相关酶及蛋白在海马区的表达而发挥作用。 Objective To establish rat models of AD to explore the pre-disposal on the rat models of senile dementia Wnt signal transduction pathway,and reveal the mechanism of acupuncture treatment of AD.Methods All the rats were divided into normal,sham,model,pre-moxibustion,pre-electro-acupuncture,pre-electro-acupuncture and moxibustion groups at random. Results Wnt1 gene singal expression was observed to weaken abnormally in model rats. There were significant difference between the model group and sham,normal groups (P0.01),while the Wnt1 singal were reinforced in pre-electro-acupuncture,pre-moxibustion and pre-electro-acupuncture groups in different degree. Conclusions The adjusting mechanism of acupuncture to prevent and cure dementia is likely to regulate the Wnt signal transduction pathways and affect the relevant enzymes and proteins' expression of learning and memory in the hippocampus.
