脂质体介导TIMP-2转染人成釉细胞瘤细胞的时效性分析 被引量:1

摘要 用阳性脂质体LipofectamineTM2000将不同浓度(1、2、3μg)的TIPM-2真核表达载体pcDNA3.1(+)/GFP-TIMP-2转染至人AM细胞,倒置相差荧光显微镜观察转染情况,流式细胞仪测定转染效率。转染24 h后细胞生长良好,72 h后细胞体积增大,细胞排列变疏松,部分细胞出现核溶解。1μg组的转染率与2、3μg组的转染率在各个时段都有明显的差别,差异有显著性。3μg组的转染效率最高,72 h达到53.9%,但3μg组与2μg组的转染率差异无显著性。TIMP-2可成功转染至体外培养人成釉细胞瘤细胞,2μg为最优的转染浓度。 After the pcDNA3.1(+)/GFP-TIMP-2 was transfected into cultured human ameloblastoma cells,Flow Cytometry(FCM) and Microscopy were performed to evaluate the effect of transfection.At 72 h,the rate of transfection was 53.9% in 3 μg group.The green fluorescence was found in plasm(observed with fluo-microwave).pcDNA3.1(+)/GFP-TIMP-2 was successfully transfected into cultured human ameloblastoma cells.2 μg TIMP-2 may be the best for transfecting.
