摘要
目的:探讨EB病毒编码的LMP1在体内鼻咽癌细胞中,是否可能通过影响鼻咽癌细胞增殖和凋亡的生物学过程,而在鼻咽癌的发展演进过程中起作用。方法:采用TUNEL法原位检测癌细胞的调亡程度,用免疫组化LSAB法检测LMP1、PCNA、p53和bcl-2蛋白表达Z分析LMP1的表达与癌细胞增殖、凋亡以及与p53、bcl-2表达的相关性。结果:LMP1的表达与否,与鼻咽癌细胞增殖活性的差异无显著意义,而与癌细胞凋亡程度之间的差异有显著意义,其表达与癌细胞凋亡程度呈正相关;LPM1的表达与p53和bcl-2的表达无相关性。结论:提示LMP1在体内鼻咽癌组织中可能有促使癌细胞凋亡的生物学作用,其机制可能是通过激活细胞毒性T淋巴细胞,增强后者对癌细胞的杀伤作用。LMP1对癌细胞增殖和凋亡的作用可能并不受p53及bcl-2表达的影响。
Purpose: To explore the relationship between Epstein-Barr encoded LMP-1 and proliferative activity as well as apoptosis in human nasopharyngeal carcinoma cells.Methods:Apototic carcinoma cells were identified by the terminal deoxynucletidyl transferase-medicated dUTP-biotin nick end labeling(TUNEL) method. LMP-1 .PCNA、p53 and bcl-2 were detected by immunohistochemistry. Result: Apoptosis of carcinoma cells was increased in LMP-1 positive cases and reduced in LMP-1 negative cases. There is no statistically significant difference between the expression of LMP-1 and proliferative activity of carcinoma cells. The expression of LMP-1、p53 protein accumulation and bcl-2 expression were not correlated with each other.ConcluSion:It is likely that LMP-1 plays a role in enhancing apoptosis of nasopharyngeal carcinoma cells in vivo. One of the mechanisms is probably stimulation of cytotoxic T-cells which induce apoptosis. The effect on proliferative activity and apoptosis of LMP-1 is probably not regulated directly by bcl-2 and p53.
