摘要
目的:探讨益气活血方(脑络欣通)和补肾生髓方对脑缺血再灌注大鼠缺血侧额顶叶皮质Math-1、Mash-1蛋白表达的影响。方法:线栓法制作局灶性脑缺血再灌注大鼠模型。随机分为假手术组、模型组、益气活血方组和补肾生髓方组。脑缺血2小时,分别再灌注1、2、3周,采用免疫组化SABC法检测Math-1、Mash-1蛋白表达。结果:在缺血区额顶叶皮质,假手术组胞浆可见少量棕黄色染色Math-1和Mash-1表达阳性细胞;再灌注各时间点模型组Math-1、Mash-1表达阳性面积单位、平均吸收光密度值较假手术组明显增加(P<0.05或P<0.01)。再灌注1、2周时,益气活血方组的Math-1、Mash-1表达阳性面积单位与平均光密度值较模型组显著增加(P<0.05或P<0.01);补肾生髓方组Math-1(2、3周)、Mash-1(1、2、3周)表达的阳性面积单位或平均光密度值较模型组显著增加(P<0.05或P<0.01)。再灌注1周时,益气活血方组Math-1表达的阳性面积单位和平均光密度值较补肾生髓方组显著增加(P<0.05或P<0.01);再灌注2、3周时,补肾生髓方组Math-1表达的阳性面积单位和平均光密度值较益气活血方组显著增加(P<0.05或P<0.01);再灌注2周时,补肾生髓方组Mash-1表达的阳性面积单位,较益气活血方组显著增加(P<0.01)。结论:两种中药复方均能通过增强缺血侧脑组织额顶叶皮质Math-1和Mash-1的表达,促进局灶性脑缺血后内源性神经干细胞的增殖分化。
Objective:To discuss the effects of two kinds of Chinese medicine prescriptions (yiqi huoxue prescription and bushen shengsui prescription) on protein math-1 and mash-1 expression in brain tissue of rats with focal cerebral ischemia-reperfusion injury.Methods:The models of focal cerebral ischemia-reperfusion injury were made by intrauminal thread occlusion.Rats were randomly divided into four groups:sham-operation,model,yiqi huoxue prescription and bushen shengsui prescription groups,each group was given the corresponding medicine,cerebral ischemia for 2 hours,respectively reperfused for 1,2,3 weeks.Protein math-1 and mash-1 expression in brain tissue of rats were determined by immunohistochemical SABC method.Results:The protein math-1 and mash-1 expression in brain tissue of rats in sham-operation group were slight,the expressions of the two protein in model group were more than those in the sham-operation group after reperfused 1,2,3 weeks;the expression of the two protein in the yiqi huoxue prescription group were significantly more than those in the model group after reperfused 1,2 weeks,the expression of the two protein in the bushen shengsui prescription group obviously increased after reperfused 1,2,3 weeks;The protein math-1 and mash-1 expression in brain tissue of rats in yiqi huoxue prescription group were more than those in bushen shensui prescription group in reperfusion 1week,the expression of the two protein in bushen shensui prescription group were obviously more than those in yiqi huoxue prescription group in reperfusion 2,3 weeks.Conclusion:The two kinds of Chinese herbs prescriptions could promote the proliferation and differentiation of endogenous neural stem cell by enhancing the protein math-1 and mash-1 expression in brain tissue of focal cerebral ischemia-reperfusion injury.
