摘要
目的探讨壳聚糖-纳米羟基磷灰石复合材料(CTS-nHA)修复兔骨缺损时BMP-2在其局部的表达。方法新西兰大白兔18只,随机分为CTS-nHA组、纳米羟基磷灰石(nHA)组、空白组。于兔双侧后肢胫骨近端各制造2个约0.5cm大小的缺损,分别植入相应的材料或不植入任何材料,于术后2、4、6、10周取标本进行骨生长因子BMP-2和组织学的观察。结果可直接观察到CTS-nHA表面有较强的BMP-2表达,CTS-nHA组新生骨组织中BMP-2表达的量比nHA组高峰提前,在观察的各时间段CTS-nHA组新生的骨组织均明显高于nHA组和空白组(P<0.05),至第10周时CTS-nHA组已形成较成熟的板层骨组织,而nHA组网状编织骨间夹杂有较多的骨痂基质。结论 CTS-nHA较强的成骨活性可能与其在体内具有诱导成骨活性有关。
Objective To explore the effect of expression of BMP-2 in chitosan-nanohydroxyapatite (CTS-nHA) composite materials repair rabbits tibial bone defect.Methods 18 adult New Zealand rabbits were randomly divided into 3 groups,CTS-nHA group,nHA group,control group.Rabbits were operated with 0.5 cm osteomoty at two tibial bone,the materials were embeded into the defect position respectively,after 2,4,6 and 10 weeks post-operation,the samples were harvested and observed with pathogical and immunohistochemistry technical.Results We had observed there was plenty BMP-2 expression on the surface of CTS-nHA,farthermore,the BMP-2 expression in CTS-nHA group was more abundant than HA group and control group at the early time.There was the most abundant new bonematrix migrating into CTS-nHA group at each weeks we had observed(P〈0.05).At the tenth week,in CTS-nHA group,the new bone matrix had formed lamellar bone.But in the control group there was more bone matrix.Conclusion CTS-nHA composite materials can effectively induce bone tissue regeneration which is relevant to good osteoinductive activity.
