不同浓度甲泼尼龙琥珀酸钠鼓室内给药对其内耳受体表达的影响 被引量:4

摘要 目的探讨不同浓度的甲泼尼龙琥珀酸钠(MTH)经鼓室内给药后对豚鼠内耳中糖皮质激素受体表达的影响,分析类固醇激素鼓室内给药的可能机制,为临床治疗及用药提供理论依据。方法经鼓室注射32、40、62.5g/L3种不同浓度的MTH溶液,分别于注射后1、1.5、2h断头取耳蜗,用糖皮质激素单克隆抗体,采用免疫组化SP方法检测糖皮质激素受体(GR)在豚鼠耳蜗中的表达。结果不同浓度MTH鼓室内给药后糖皮质激素受体在豚鼠耳蜗各回均有不同程度的染色,主要阳性部位是螺旋韧带、血管纹、螺旋器和螺旋神经节。与正常组相比不同浓度的MTH鼓室内给药后糖皮质激素受体在耳蜗各阳性部位的表达均明显增强,且同一时间点浓度越高其受体表达越强,而同一浓度又以1.5h时间点表达最强,即在1.5h取材点、62.5g/LMTH鼓室内给药后糖皮质激素受体在豚鼠内耳中表达强度最大,其中在螺旋韧带和血管纹的表达增强最大,有统计学意义(P<0.05)。3种浓度的MTH鼓室内给药后2h取材时间点的ABR波形Ⅲ波阈值较对照组差异均无统计学意义(P>0.05)。结论 MTH鼓室内给药可使糖皮质激素受体在豚鼠耳蜗中的表达增强。且其表达强度和时间及浓度有关。 Objective To investigate the effect of MTH of different density to the glucocorticoid receptor′s express on cavia cobaya′s auris interna injected through the cavum tympani.Then to analyze the possible mechanism of the administration through the cavum tympani.Methods MTH of three kind of densitie(32,40 and 62.5 g/L)was injected to the cavum tympani,we operated decapitation to get the cochlea in 1,1.5 and 2 h after injected.The expression of glucocorticoid receptor in cavia cobaya′s auris interna was detected by immunohistochemistry with monoclonal antibody of glucocorticoid receptor.Results Glucocorticoid receptor got tinctured in each part of the cochlea after injected MTH of different density through the cavum tympani.The positive espression part was in ligament of cochlea,psalterial cord,acoustic organ and ganglion spirale.Compared with normal group,glucocorticoid receptors expression got stronger in each part of the cochlea after injected MTH of different density through the cavum tympani.The intension of the receptors expression got stronger as the density of the MTH changed higher at the same time point.Under the condition of the same density,the strongest expression appeared at 1.5 h after injected.That suggested the strongest expression of the glucocorticoid receptor happened at 1.5 h with injecting MTH of 62.5 g/L,especially in spiral and psalterial cord.The difference of the receptors expression had statistical significance(P〈0.05).The Ⅲ wave′s threshold of ABR had no statistically significan compared with the control group at 2 h after injected three different density through the cavum tympani(P〈0.05).Conclusion The expression of glucocorticoid receptor in cavia cobaya′s auris interna can get stronger through injecting MTH in cavum tympani.And the intension of expression has relative with the time and density.
