摘要
为观察经转化生长因子β1(TGF-β1)诱导的新生儿脐带血Treg中Foxp3和IL-10表达特点,探讨TGF-β1在诱导产生的新生儿脐带血Treg中的作用。将脐带血单个核细胞用CD3mAb激活后,加IL-2培养扩增,同时加入不同浓度的TGF-β1干预,扩增第4天分别用流式细胞术和RT-PCR技术检测CD4+CD25+TregFoxp3蛋白和Foxp3 mRNA的表达,用ELISA法检测扩增细胞培养上清液中IL-10水平。结果显示,TGF-β1诱导下脐带血CD4+CD25+TregFoxp3蛋白和Foxp3 mRNA的表达明显增加,培养上清中IL-10含量也明显增加。表明,TGF-β1在体外能够促进脐带血CD4+CD25+Treg的Foxp3的表达和IL-10分泌。
To observe the expression of Foxp3 and IL-10 on CD4+CD25+ Treg induced by transforming growth factor-β1 (TGF-β1) in neonatal cord blood,cord blood mononuclear cells(CBMC) were stimulated with CD3 mAb and cultured in IL-2 containing medium with different concentrations of TGF-β1. The expressions of Foxp3 protein and Foxp3 gene mRNA in CD4+CD25+ Treg were detected by flow cytometry and RT-PCR assay,respectively,and the level of IL-10 in supernatant from cultured CBMC was detected by enzyme linked immunosorbent assay (ELISA). It was demonstrated that the expression of Foxp3 protein and mRNA in CD4+CD25+ Treg and IL-10 level in supernatant of cultured CBMC increased significantly in TGF-β1 treated group compared to control group. It is concluded that TGF-β1 up-regulates Foxp3 expression of the CD4+CD25+Treg and increases the secretion of IL-10 in cord blood in vitro.
