加热对肺癌A549细胞生长和细胞周期影响的实验研究 被引量:3

Influence of hyperthermia on growth and cell cycle in lung cancer A549 cells

摘要 目的:研究加热对肺癌A549细胞生长和细胞周期的影响。方法:体外复苏与培养肺癌A549细胞;对照组37℃下培养,实验组按不同时间、不同温度分组水浴加热肺癌A549细胞后培养24h,采用四甲基偶氮唑盐比色法(MTT)检测细胞增殖抑制率;采用流式细胞术观察热处理后的肺癌A549细胞周期分布的变化。结果:MTT实验提示38℃加热对A549细胞生长无明显抑制(P>0.05),40℃、42℃加热对A549细胞生长有抑制作用(P<0.05),并且该作用与加热的温度和时间成正相关;流式细胞术显示,加热能改变A549细胞周期分布,使S期细胞明显增多(P<0.01),G0/G1期和G2/M期细胞明显减少,P<0.05。结论:加热可抑制A549细胞生长,改变肿瘤细胞周期分布并可能因此增强化疗疗效。 OBJECTIVE:To investigate the influence of hyperthermia on the growth and cell cycle in lung cancer A549 cells by different temperature and heating time thereby providing the academic evidence in hyperthermia and thermochemotherapy for lung cancer.METHODS:The A549 cells were resuscitated and caltured in vitro.Control group:A549 cells were cultivated at 37 ℃.Experimental group:A549 cells were cultivated 24 hour after heated by different temperature and heating time.MTT assay was adopt to evaluate the inhibitory effect of A549 cells.The flow cytometry(FCM)was adopted to observe the influence on the cell cycle.RESULTS:According to the data of MTT assay,the 38 ℃ heating did not inhibit the proliferation of A549 cells(P〉0.05),40 ℃ and 42 ℃ heating inhibited the proliferation of A549 cells(P〈0.05),and there was a positive correlation between the heating dose(temperature and time)and inhibitory effect.The FCM showed that the cell cycle of A549 cells was arrested in S phrase(P〈0.01),the number of A549 cells in G0/G1 phase and G2/M phase was decreased obviously after heating(P〈0.05).CONCLUSION:Hyperthermia could inhibit the growth of A549 cells and change the distribution of cell cycle which can enhance the effect of chemotherapy.
