缺血再灌注过程中MMP-9和IL-8的变化及其相关性研究

摘要 目的探讨MMP-9和IL-8在缺血再灌注过程中的变化及其相关性。方法采用酶联免疫吸附法(ELISA)检测46例大白兔(伪手术组7只,缺血再灌注2h组、4h组及6h组分别为10、10、9只,心肌梗死组10只)12h~7d血浆中MMP-9及IL-8的含量。结果缺血再灌注组MMP-9从12h时逐渐升高,第3天达高峰,4~7天显著下降。缺血再灌注组IL-8从12h开始升高,第2天达高峰,以后显著下降,第5天达正常水平。12h~7d缺血再灌注组MMP-9明显高于伪手术组(P<0.05),但明显低于心肌梗死组明显(P<0.05);缺血再灌注4h组明显高于缺血再灌注2h组(P<0.05),明显低于缺血再灌注6h组(P<0.05)。12h~3d缺血再灌注.IL-8明显高于伪手术组(P<0.05),明显低于心肌梗死组(P<0.05);MMP-9和IL-8在缺血再灌注4h组明显高于缺血再灌注2h组(P<0.05),明显低于缺血再灌注6h组(P<0.05)。结论血浆MMP-9及IL-8水平可预测急性心肌梗死的严重性,在3天内缺血再灌注组血管再通后血浆MMP-9与IL-8显著正相关,血浆IL-8在缺血再灌注过程中可进一步促进血浆MMP-9水平升高。 Objective To explore the changes of matrix metallop-- roteinases--9(MMP--9) and interleukin--8 in the process of isehemical reperfusion and their relative research. Methods The plasma MMP--9 and IL--8 level were measured in 46 rabbits (sham operation group (n ----- 7), Ischemic reperfu- sion group 2h group (n = 10), 4h group (n = 10) and 6h group (n = 9) and myocardial infarction group (n ----- 10) 12h^7days by enzyme linked immunosorbent assay (EI. ISA). Results MMP--9 gradually be- gan to rise after 12h in ischemie reperfusion group, reached peak in third day, and then gradually de- clined. IL--8 gradually began to rise after 12h in ischemic reperfusion group, reached peak in second day, and then gradually declined, but recovered normal level in fifth day. MMP--9 of isehemic reperfu- sion group were dramatically higher than sham group's in 12h~ 7d (P ~ 0.05). and significant lower than myoeardial infarction group (P 〈 0. 05). IL--8 of ischemic reperfusion group were markedly higher than sham group's in 12h^3d (P 〈 0. 05), and remarkably lower than myocardial infarction group (P 0.05). MMP--9 and IL--8 of ischemic reperfusion 4h group were significant higher than isehemic reper- fusion 2h group (P 〈 0.05), and remarkably lower than ischemic reperfusion 6h group (P 〈 0.05). Conclusions Blood plasma level of MMP--9 and IL--8 can pretest severity of acute myocardial infarc- tion. Blood plasma MMP--9 level are closely correlated with blood plasma IL--8 level in ischemia reper- fusion group during 3 days. Blood plasma IL--8 can raise blood plasma MMP--9 level in the process of ischemic reperfusion.
