丹酚酸B对缺血再灌注大鼠肾脏缝隙连接蛋白43表达的影响

摘要 目的研究丹酚酸B对缺血再灌注大鼠肾脏缝隙连接蛋白43(Cx43)表达的影响,探讨Cx43在丹酚酸B抗肾缺血再灌注损伤(RIRI)中的作用及其意义。方法 SD大鼠30只,分3组,每组10只,假手术(Sham)组、缺血再灌注(IR)组、丹酚酸B(Sal-B)组,于再灌注2h后检测肾组织匀浆丙二醛(MDA)、超氧化物歧化酶(SOD);肾组织切片HE染色行中性粒细胞(PMN)计数及Paller评分,量化肾损害程度;Western-blot定量分析Cx43蛋白在肾组织中的表达。结果肾组织中MDA水平:Sal-B组明显低于IR组;SOD水平:Sal-B组明显高于IR组;PMN计数及Paller评分:Sal-B组明显低于IR组;Cx43蛋白表达:IR组明显低于Sham组,Sal-B组明显高于IR组。结论丹酚酸B提高Cx43在肾组织中的表达,减轻肾缺血再灌注损伤。 [ Objective] To observe the effect of salvianolic acid B(Sal-B) on connexin43(Cx43) expression after renal ischemie reperfusion OR) in rats, and investigate the effect of Cx43 in the protective mechanism of Sal-B on renal isehemic repeffusion injury. [Methods] 30 SD rats were divided into three groups randomly (n=10): sham group , ischemie reperfusion group 0R-group), salvianolic acid B group (Sal-B-group). The kidney underwent 60- minute isehemic and 2-hour reperfusion. The value of MDA and SOD were examined. The changes in renal pathological morphology were assessed by PMNs and Paller count. The expression levels of Cx43 protein of kidney was examined by western blot. [Results] Renal MDA: The values in Sal-B- group were significantly lower than those in IR-group; SOD: the values in Sal-B- group were signifieantly higher than those in IR-group. The numbers of PMNs and Paller's count: The values in Sal-B-group were significantly lower than those in IR-group. Compared with Sham-group, expression of Cx43 in IR-group was significantly decreased. The protein expression in Sal-B- group was significantly increased as compared with that of IR-group. [ Conclusion ] Salvianolie acid B could preteet kidney from isehemie reperfusion injury. The mechanism might be that salvianolie acid B increases the protein expression of Cx43 in renal tissue.
