Genistein抑制卵巢癌细胞系SKOV3增殖和侵袭能力的体外研究

摘要 目的探讨Genistein对卵巢癌细胞株SKOV3增殖和侵袭能力的影响。方法用不同浓度的Genistein处理SKOV3细胞,MTT比色法、克隆形成实验测定其对细胞增长的抑制情况,细胞黏附实验、Transwell小室观察其对细胞黏附、运动和侵袭能力的影响。结果 Genistein抑制SKOV3细胞的生长,并呈剂量依赖性(P<0.05);其对SKOV3细胞的运动和侵袭能力均有明显抑制作用,其抑制率呈剂量效应曲线(P<0.05)。结论 Genistein对卵巢癌细胞株SKOV3的增殖和侵袭能力均有明显抑制作用。 [Objective] To examine the inhibitory effect of genistein on invasion and proliferation of human ovarian cancer cell line SKOV3. [Methods] The SKOV3 cell line was exposed to various eoneentration of genistein. Methyl thiazolyl tetrazolium (MTT) assay and eolon formation assay were used to observe the growth inhibition rate of SKOV3 eeU line. Cell adhesion test and transwell assay were used to evaluate the adhesive, migration and invasion ability of SKOV3 cell line. [.Results] Genistein could inhibit the growth of SKOV3 cell line, and this inhibition was in a dose-dependent pattern (P 〈0.05). Genistein could reduce abilities of migration and invasion in SKOV3 cell line in a dose-dependent pattern significantly (P 〈0.05). [ Conclusions ] Genistein can inhibit the proliferation and invasion ability of ovarian cancer in vitro.
