摘要
目的:研究大蝎子草水层部位对脂多糖(LPS)诱导大鼠腹腔巨噬细胞诱导型一氧化氮合酶(iNOS)表达、一氧化氮(NO)与肿瘤坏死因子α(TNF-α)分泌的影响。方法:建立体外培养大鼠腹腔巨噬细胞,以LPS为炎症刺激剂,免疫细胞化学研究大蝎子草水层部位不同剂量对LPS诱导大鼠腹腔巨噬细胞iNOS表达,Griess法分析培养液NO含量,间接MTT法分析培养液TNF-α含量。结果:LPS刺激大鼠腹腔巨噬细胞后,其iNOS表达显著增加,培养液中NO和TNF-α含量显著增加,大蝎子草水层部位对上述改变具有显著的抑制作用。结论:大蝎子草水层部位对细胞水平的炎症反应具有一定的抑制作用。
Objective:To investigate the inhibited effects of of Herba Girardinia water extract on inducible nitric oxide synthase (iNOS) expression in macrophages,and nitric oxide (NO) and tumor necrosis factor-alpha (TNF-α) excretion in macrophages induced by lipopolysaccharide (LPS).Methods:The model was reproduced by LPS stimulating macrophages in culture in vitro.Induced NOS was detected by immuocytochemistry method,NO contents in medium were assayed by Griess method,and the TNF-α contents in medium were measured by indirect MTT method.Resutls:Induced NOS expression of macrophages,NO and TNF-α contents in medium of macrophages are significantly increased after the stimulation by LPS.Herba Girardinia water extract can remarkable ameliorate the indexes.Conclusion:Herba Girardinia water extract inhibits the inflammatory reaction at cell level.
