益肾I方对PCOS大鼠胰岛素敏感性、细胞因子及氧化应激的作用 被引量:1

Effects of Yishenyifang decoction on insulin sensitivity,proinflammatory cytokines and oxidative stress~free radical injury in rats with PCOS

摘要 目的:观察益肾I方对多囊卵巢综合征(PCOS)大鼠胰岛素抵抗及其相关的促炎细胞因子和脂肪细胞因子以及氧化应激反应的干预作用。方法:采用十一酸睾酮加绒促性素(HCG)建立PCOS大鼠模型,连续口服给予罗格列酮10mg/kg、益肾Ⅰ方25g/kg,12.5g/kg,6.25g/kg3个剂量6周,观察大鼠体重、空腹血糖(FBG)、口服葡萄糖后2小时血糖(OGTT-2hBG)、空腹胰岛素(FIns)、高敏C反应蛋白(hs-CRP)、肿瘤坏死因子-α(TNF-α)、瘦素(leptin,L)、脂联素(adiponectin,A)、抵抗素(resistin,R)、睾酮(testosterone,T)、丙二醛(MDA)、一氧化氮(NO)、以及超氧化物歧化酶(SOD)和诱导型一氧化氮合酶(iNOS)-一氧化氮合酶(NOS)活性等的含量,并计算胰岛素敏感指数(insulinsensitivityindex,ISI)的变化。结果:PCOS大鼠FBG,OGTT-2hBG,Finshs-CRP,TNF-α,leptin,resistin,T,胰岛素水平以及iNOS-NOS活性水平明显升高,而ISI,SOD和adiponectin含量显著降低,与空白对照组比较差异有高度显著性(P<0.01);罗格列酮和益肾I方三个剂量组均能不同程度地改善PCOS大鼠的上述病理改变(P<0.01~0.05)。结论:罗格列酮及益肾Ⅰ方均能不同程度地改善PCOS大鼠高胰岛素血症、高雄激素血症、胰岛素抵抗(IR)和血糖代谢异常,下调促炎细胞因子和脂肪细胞因子表达,以及对抗氧化应激-自由基损伤,并提高抗氧化能力。 Objective:To investigate intervening effects of Yishenyifang decoction on insulin resistance and it closely related to blood glucose,insulin sensitivity,cytokines and oxidative stress~free radical injury in rats with polycystic ovary syndrome (PCOS).Method:The model of PCOS rats induced by testosterone undecanoate and human chorionic gonadotropin (HCG),afterwards rosiglitazone 10mg/kg,Yishenyifang decoction 25g/kg,12.5g/kg,6.25g/kg were taken by intragastric administration for 6 weeks.At the end of the experiment,the contents or activities of fasting blood~glucose (FBG),2~hours blood glucose after oral glucose tolerance test(OGTT~2hBG),insulin(Ins),high sensitivity C~reactive protein(hs~CRP),tumor necrosis factor (TNF) ~α,leptin(L),adiponectin(A),resistin(R),testosterone (T) malondialdehyde (MDA),nitric oxide (NO),as well as superoxide dismutase (SOD) and inducible nitric oxide synthase (iNOS) ~ nitric oxide synthase (NOS) activity in were determined,and the changes of insulin sensitivity index (ISI) were counted.Result:Compared with the normal group,the concentrations or activities of FBG,OGTT~2hBG,Ins,hs~CRP,TNF~α,L,R,T,insulin levels,and iNOS~NOS activity in were increased obviously(P0.01),but ISI,A were decreased significantly (P0.01).Rosiglitazone and Yishenyifang decoction could improved the pathologic changes in PCOS rats (P0.01or P0.05).Conclusion:Rosiglitazone and Yishenyifang decoction can improve the pathologic changes in PCOS rats on IR,dysglycemia,hyperinsulinemia,hyperandrogenemia,and downregulating proinflammatory cytokines and adipocytokines by which degrees differ,as well as the fight against oxidative stress ~ free radical damage and enhance antioxidant capacity.
