摘要
目的观察阿托伐他汀对单侧输尿管结扎大鼠肾间质纤维化的调节作用。方法采用单侧输尿管结扎术致肾间质纤维化大鼠模型,随机分为假手术组、模型组、治疗组。术后第3、7、14天处死,采取空腹静脉血监测总胆固醇,经HE、Masson染色及免疫组化观察肾间质损伤及肝细胞生长因子(HGF)的表达。结果 3组胆固醇浓度差异无统计学意义(P>0.05),模型组肾间质损伤明显高于假手术组(P<0.05),而治疗组HGF表达高于模型组(P<0.05),肾间质损伤低于模型组(P<0.05)。结论阿托伐他汀通过非降脂途径延缓单侧输尿管梗阻大鼠(UUO)所致大鼠肾间质纤维化程度,部分依赖于增强HGF表达。
Objective To investigate the protective effect of atorvastatin on renal interstitial fibrosis of unilateral obstruction(UUO) in rats.Methods The rats models of renal interstitial fibrosis were made by unilateral urethral ligation,the rats were randomly divided into three groups,sham operation group,model group and treatment group.The total cholesterol was detected,the histological changes in obstructed kidney were observed by HE stain and Masson stain.The changes of expression of HGF were analyzed by routine immunohistochemical method.Results As compared with those in sham operation group,there were no significant differences in the levels of TC between model group and treatment group(P〉 0.05).The renal interstitial injury in model group was significantly severer than that of sham operation group(P〈 0.05),however,which in treatment group was obviously slighter than that of model group(P〈 0.05),and the levels of HGF in treatment group was significantly higher than that in model group(P 〈0.05).Conclusion The atorvastatin can relieve renal interstitial fibrosis caused by UUO in rats by increasing HGF protein expression.
