β-catenin,cyclin D1和ERα在乳腺癌组织中的表达 被引量:3

Expression of β-catenin,Cyclin D1 and ERα in Breast Cancer

摘要 目的检测β-连接蛋白(β-catenin)、周期素D1(cyclin D1)和雌激素受体α(ERα)在乳腺癌中的表达,探讨其在乳癌发病机制中的作用。方法随机选取42例乳腺增生,58例乳腺癌病人的乳腺组织以及乳腺癌灶周围正常乳腺组织,分别应用免疫组化SP法检测β-catenin,cyclin D1的表达,应用Western-blot法测ERα的表达。结果乳腺癌组织β-catenin呈胞质和/或胞核异位表达,其表达率为65.5%,显著高于乳腺增生和正常乳腺组织(42.9%、35.7%,P<0.05)。乳腺癌cyclin D1和ERα阳性率分别为74.1%、70.7%,均显著高于乳腺增生和正常乳腺组织(45.2%、36.5%;33.3%、29.5%;P均<0.01)。乳腺癌中β-catenin与cyclin D1和ERα表达分别呈明显正相关(r=0.32、0.57,P<0.05、P<0.01)。结论乳腺癌中ERα、cyclin D1和β-catenin均高表达,且呈正相关,提示三者在乳腺癌发病中具有协同作用。 Objective To test the expression of β-catenin,cyclin D1 and ERα in breast cancer,and to explore the role of β-catenin,cyclin D1 and ERα in the pathogenesis of breast cancer.Methods 42 tissue samples of mammary gland hyperplasia and 58 samples of breast cancer tissue and normal tissue near the breast cacer were randomly selected for the study.The expression of β-catenin and cyclin D1 was determined by immunohistochemistry SP test.And the expression of ERα was detected by Western blot analysis test.Results The ectopic expression rate of cytoplasm and/or nucleus of β-catenin in breast cancer(65.5%)was significantly higher than those in mammary gland hyperplasia(42.9%)and normal breast tissue(35.7%),(P〈0.05).The positive rates of cyclin D1 and ERα in breast cancer were 74.1% and 70.7%,respectively;which were significantly higher than those in mammary gland hyperplasia(45.2%,36.5%)and normal breast tissue(33.3%,29.5%),(all P〈0.01).The expression of cyclin D1 and ERα was positively correlated with that of β-catenin in breast cancer(r=0.32,r=0.57;P〈0.05,P〈0.01).Conclusions β-catenin,cyclin D1 and ERα are highly expressed in breast cancer,and moreover,the expression of cyclin D1 and ERα is positively associated with that of β-catenin.The results suggest that β-catenin,cyclin D1 and ERα have a synergistic effect on the pathogenesis of breast cancer.
