摘要
目的研究Cyr61蛋白在人结肠癌血管生成中的作用及临床意义。方法采用免疫组织化学染色法检测人结肠癌肿瘤组织中Cyr61、VEGF和VEGFR2的表达情况,用CD34单克隆抗体标记血管内皮细胞,并计算肿瘤微血管密度(MVD),用SPSS10.0软件分析Cyr61与VEGF、VEGFR2及肿瘤MVD的相关性。结果 (1)Cyr61、VEGF及VEGFR2在50例人结肠癌肿瘤组织中阳性表达率分别为90.0%、70.0%和60.0%,明显高于正常结肠组织(P=0.002,P=0.000,P=0.001);(2)Cyr61、VEGFR和VEGFR2蛋白阳性表达组微血管密度较阴性表达组高(P<0.05);(3)与临床Ⅰ~Ⅱ期或无淋巴结转移病例比较,临床Ⅲ~Ⅳ期和有淋巴结转移的病例Cyr61、VEGF和VEGFR2蛋白表达率较高(P<0.05),Cyr61表达与临床分期和淋巴结转移呈正相关(r=0.333,r=0.308,P=0.018,P=0.030);(4)Cyr61蛋白表达与VEGF和VEGFR2的表达呈正相关(r=0.364,r=0.408,P=0.009,P=0.003)。结论 Cyr61可促进结肠癌肿瘤血管生成,其作用机制可能与VEGF/VEGFR2通路有关。
Objective To study the angiogenesis effect of Cyr61 and its clinical significance in human colon cancer. Methods Expression of Cyr61, VEGF and VEGFR2 Proteins were detected by immunohistochemistry. Microvessel density (MVD) was obtained after staining of microvessels using CD34 antibody. SPSS10.0 statistical software was used to analyze the correlations between the expression of Cyr61,VEGF and VEGFR2 Proteins and MVD in tumor tissues of colon cancer. Results (1) Expression of Cyr61 ,VEGF and VEGFR2 on the tumor cells of human colon cancer were positive in 90.0%, 70.0% and 60.0%, respectively. Furthermore,there was a statistical higher positive rate compared with that of normal colon tissues (P=0.002,P--0.000 and P=0.001,respectively). (2) These cases with positive expression of Cyr61, VEGFR and VEGFR2 proteins had a higher MVD in tumor tissues, compared to that of by comparing to that of negative expression of these proteins in human colon cancer(P〈O.05). (3) Compared to stage Ⅰ~Ⅱ or without lymph node metastasis group, these cases in stage Ⅲ-Ⅳ and with lymph node metastasis had a higher positive rate of Cyr61, VEGF and VEGFR2(P〈0.05 ). And the expression of Cyr61 was positively correlated to clinical stage and lymph node metastasis(r=0.333 and r=0.308,P=0.018 and P=0.030, respectively). (4) Expression of Cyr61 was positively correlated to the expression of VEGF and VEGFR2(r=0.364 and r=0.408,P=0.009 and P=0.003, respectively). Conclusion Cyr61 may promote the angiogenesis in tumor tissues of human colon cancer through VEGF/VEGFR2 signaling Pathway.
