摘要
采用毛细管电泳分析手段,用校正峰面积归一化法同时测定了本实验室购得的α-Lac、β-LgA及β-LgB三个蛋白参考物的纯度,其值分别为75.4%、84.5%和76.9%,相对标准偏差分别为1.6%、0.7%及1.4%。所得结果与应用十二烷基硫酸钠-聚丙烯酰胺平板凝胶电泳法的测定结果进行了比较,并讨论了造成二者差异的原因。结果表明毛细管电泳法是分析此类蛋白的有效潜在手段。
The purity of three proteins,i.e.,α-lactalbumin(α-Lac),β-lactoglobulin A(β-LgA) and β-lactoglobulin B(β-LgB),were simultaneously analyzed and quantified by normalized /ected peak area method with high-performance capillary electrophoresis(HPCE).The values for α-Lac,β-LgA and β-LgB determined by this method were 75.4%,84.5% and 76.9%,with relative standard deviation values(RSDs) of 1.6%,0.7% and 1.4%,respectively.The results were then compared with those obtained by sodium dodecyl sulfate-polyacrylamide gel elelctrphoresis(SDS-PAGE) method.The reasons for differences between the two results were discussed.HPCE method has proved to be a powerful tool for purity analysis of such proteins.
