丁胺卡那霉素地塞米松复方脂质体的眼毒性研究

摘要 为探讨治疗细菌性眼内炎的新型制剂丁胺卡那霉素地塞米松复方脂质体的视网膜毒性,将20只新西兰白兔随机分为四组,第一、二、三组分别于玻璃体内注射不同剂量的丁卡地塞复方脂质体,第四组玻璃体内注入注射用水作对照。术后行裂隙灯、检眼镜活体检查,并行ERG检查和视网膜透射电镜检查。结果:含丁卡600μg、地塞米松525μg的0.15ml复方脂质体对视网膜无明显毒性,而含丁卡800μg、地塞米松700μg的复方脂质体则显示毒性反应。结论:丁卡地塞复方脂质体较游离药物毒性小、作用时间长。 Objective:To study the retinal toxicity of a new preparation of liposome encapsulated amikacin and dexamethasone for treatment of bacterial endophthalmitis.Methods:20 rabbits were randomly into 4 groups.Rabbits in group 1,2,3 received intravitreal injection of the compound liposome at different dosage respectively.Rabbits in group 4 as control received intravitreal injection of sterilized water.After intravitreal injections,the eyes were examined by ophthalmoscopy,slit lamp microscope and some also by transmission electrone microscope.to all eyes,ERG were recorded.Results:No evidence of toxicity was found in retinas after intravitreal injections of the compound liposome 0 15ml (contained amikacin 600 μg and dexamethasone 525μg),but intravitreal injections of the compound liposome 0 2ml (contained amikacin 800μg and dexamethasone 700μg) induced toxic effects on the retinas.Conclusion:The liposome encapsulated amikacin and dexamethasone can reduce toxicity of the two drugs and prolonge their half life in vitreous.So this compound liposome may be a effective preparation in the treatment of endophthalmitis.
