多药耐药基因1C3435T多态性对汉族儿童外周血单个核细胞mRNA表达的影响 被引量:1

摘要 目的探讨多药耐药基因1(multidrug resistance gene 1,MDR1)C3435T多态性与健康汉族儿童外周血单个核细胞中mRNA表达的关系。方法以130例(男性83例,女性47例)无亲缘关系的中国健康汉族儿童为研究对象,采用PCR-RFLP技术检测其MDR1基因C3435T位点的基因型,实时荧光定量PCR法检测其外周血单个核细胞中MDR1mRNA表达水平。结果 130例儿童中,51例为CC型,63例为CT型,16例为TT型,基因型频率分布符合Hardy-Weinberg平衡(P>0.05);CC、CT、TT各基因型组间外周血单个核细胞MDR1 mRNA表达水平差异无统计学意义(P>0.05)。结论汉族儿童中MDR1基因C3435T多态性对外周血单个核细胞中mRNA表达无显著性影响。汉族儿童C3435T位点多态性不能作为推测MDR1表达水平的依据。 Objective To study the correlation between multidrug resistance gene 1 ( MDR1 ) C3435T polymorphism and its mRNA expression in peripheral blood mononuclear cells of healthy Han populations. Methods A total of 130 Chinese Han healthy children at a mean age of 7.62 with no consanguinity (83 males and 47 females) were included in this study. Genotypes of MDR1 C3435T polymorphism were identified by PCR-RFLP, and MDR1 mRNA expression levels in peripheral blood mononuclear cells were measured by SYBR green [ real-time polymerase chain reaction. Results The CC, CT and TT genotypes of MDR1 C3435T polymorphism were identified in 51,63, and 16 out of the 130 children, respectively. The distribution of genotypes was consistent with the Hardy-Weinberg equilibrium (P 〉 0.05). No significant difference was found in MDR1 mRNA expression levels between the CC, CT and 3T genotypes of MDR1 C3435T polymorphism in peripheral blood mononuclear cells. Conclusion MDRI C3435T polymorphism does not significantly influence its mRNA expression in peripheral blood mononuclear cells of Chinese Han children. Genotyping of this polymorphism cannot be used as the evidence for predicting the expression of MDR1 in Chinese Han children.
