摘要
目的观察雷帕霉素(Rapamycin,RAPA)对体外培养的黑色素瘤B-16细胞增殖和凋亡的影响。方法应用MTT比色法检测细胞增殖活力;采用ELISA检测细胞凋亡;倒置显微镜观察细胞形态学变化。结果 0~40 nmol·L^(-1)RAPA能使B-16细胞增殖活力降低并使其发生凋亡,且具有时-效和量-效依赖关系;显微形态表现为细胞体积缩小,粘附性能减弱。结论 RAPA能抑制黑色素B-16细胞增殖并可诱导其发生凋亡。
OBJECTIVE To investigate the effect of Rapamycin on the cell proliferation and apoptosis of cultured melanoma B-16 cells.METHODS MTT reduction assay was used to reflect the proliferation of the cells. The apoptosis were detected by ELISA. The morphological changes and apoptotic characteristics of the cells were observed with the aid of inverted microscope.Results-Rapamycin 0 - 40 nmol/L notably inhibited the growth of B-16 cells, and induced them happen apoptosis, which showed the dependence relationship between time effect and dose effect, the cell grow density got less, cell rounding and cell shrinkage could be seen under inverted microscope. CONCLUSIONS Rapamycin can remarkably inhibit proliferation and induce apoptosis of melanoma B-16 cells.
