摘要
目的探讨了血管舒缓素对血小板趋化生长因子(PDGF)诱导培养的大鼠胸主动脉平滑肌细胞增殖的影响。方法取SD大鼠胸主动脉平滑肌细胞进行培养,传至5代后,分组观察PDGF、PDGF+血管舒缓素组、PDGF+血管舒缓素+缓激肽受体拮抗剂组等。干预因素作用于血管平滑肌细胞(VSMC),对3H胸腺嘧啶(3HTdR)的掺入及原癌基因cmycmRNA表达的影响。结果1.PDGF可促进VSMC3HTdR的掺入和cmycmRNA表达,这种表达在2小时时达高峰。2.血管舒缓素可抑制PDGF诱导的VSMC3HTdR的掺入和cmycmRNA表达,这种抑制作用可被缓激肽受体拮抗剂所阻断。结论血管舒缓素可抑制PDGF诱导的VSMC增殖作用。
Objective The purpose of the present study was to explore the effect of Kallikrein on PDGF induced proliferation of rat aortic vascular smooth muscle cells (VSMC).Methods In vitro cultured rat aortic VSMC model was established. Cells of 4 6 passages were used for the experiment. At was divided into four groups: (1). Control; (2). PDGF (50ng/ml); (3). PDGF+Kallikrein(10u/ml); (4).PDGF+Kallikrein+Bradykinin receptor antagonist HOE140(10 -4 M). Proliferation of VSMC in culture was measured by 3H TdR incorporation into cell DNA. The expression of VSMC c myc mRNA was determined by using the RT PCR technique.Results (1). Compared with the control, PDGF significantly increased the 3H TdR incorporation and the expression of c myc mRNA in cultured VSMC, and the expression of c myc mRNA peaked at 2h (P<0.05). (2). The action of PDGF was inhibited by kallikrein. The effect of kallikrein on PDGF can be blocked by HOE140.Conclusion These results suggest that kallikrein can inhibit the proliferation of cultured VSMC induced by PDGF.
