摘要
目的:探讨缺氧与梭曼中毒单一或复合作用对培养大鼠心肌细胞的损伤特点和可能机制。方法:培养的大鼠心肌细胞在缺氧条件下暴露于含不同浓度梭曼的培养液中,观察细胞存活率和培养液肌酸磷酸激酶(CPK)、乳酸脱氢酶(LDH)及谷草转氨酶(GOT)等心肌酶含量变化,同时用Fura-2/AM荧光负载测定心肌细胞内游离钙浓度([Ca2+]i)变化。结果:缺氧与梭曼中毒均可导致培养的大鼠心肌细胞存活率下降,培养液中心肌酶含量升高,同时心肌细胞内游离钙浓度也明显增加;缺氧和梭曼中毒两种致伤因素同时作用对培养的大鼠心肌细胞所产生的复合效应显著大于单一因素的致伤结果。结论:缺氧可以加重梭曼对心肌细胞的毒性;
Objective: To investigate
the effects of soman intoxication or hypoxia alone and the two in combination on cultured rat
myocardial cells and the possible mechanism of myocardiocytic injury by soman intoxication
under the condition of hypoxia. Methods: The cultured rat myocardial cells were exposed to
soman solution at different concentrations under the condition of hypoxia. The changes in
contents of CPK, LDH and GOT in the culture solution were determined. Meanwhile, the
concentration of free calcium in the cells ([Ca 2+ ]i) were measured by using Fura 2 as the
fluorescent probe for intracellular Ca 2+ . Results: Both soman intoxication and hypoxia
markedly reduce the survival rate of the cultured rat myocardial cells. The contents of CPK, LDH
and GOT in the culture solution were remarkably increased after the exposure of the cells to
soman or hypoxia alone or the two in combination. The increase of [Ca 2+ ]i due to the
combination was much higher than that produced by soman intoxication or hypoxia alone.
Conclusion: Hypoxia can enhance the cytotoxic effects of soman on the myocardial cells. The
injury of the myocardial cells caused by hypoxia or soman intoxication might be related to
calcium overload in the cells.
