摘要
目的:研究缺氧所致的兔多形核白细胞-肺微血管内皮细胞粘附与缺氧时间的关系以及探讨缺氧所致粘附分子变化在其中的作用。方法:利用培养的家兔肺微血管内皮细胞及体外缺氧模型,测定家兔多形核白细胞与肺微血管内皮细胞的粘附率,运用免疫组化及流式细胞技术测定CD18与CD54的表达。结果:随着缺氧时间的延长,粘附率进行性增加,缺氧可明显加强CD18和CD54的表达,CD18和CD54单抗能明显降低粘附率的增加。结论:缺氧可直接刺激白细胞CD18和内皮细胞CD54表达。
Objective: To investigate the effects of hypoxia on adhesion of polymorphonuclear neutrophils (PMNs) to pulmonary microvascular endothelial cells (PMVECs) in rabbits. Methods: PMNs and/or cultured PMVECs were exposed to hypoxia (P O 2 =15~20 mmHg) and some of them were added with CD18 or CD54. The adhesion rate and the expression of CD18 and CD54 were determined with immunohistochemistry and flow cytometry. Results: The adhesion rate was increased by hypoxia in a time dependent manner and the expression of CD15 and CD54 was also increased by hypoxia. The two antibodies could antagonize the increase in adhesion rate. Conclusion: Hypoxia increases the expression of CD15 and CD54 which is involved in the adhesion of PMNs to PMVECs.
