摘要
目的:研究昆明山海棠对人白血病细胞HL-60HPRT位点的影响。方法:用THH水溶液对HL-60细胞进行浓度梯度染毒,不同时间点进行单细胞微孔接种,计数阳性克隆,测定接种存活率、克隆效率和突变频率。结果:THH染毒细胞的突变频率,在处理剂量3.35~20.10mg/ml范围内,具有明显的剂量-反应关系。但随着THH的浓度增高至33.50mg/ml组,由于细胞存活率太低,细胞数不够最后接种要求,不能继续进行。这一现象可能与THH的细胞毒性有关。结论:THH对HL-60细胞具有肯定的致突变性。
Objective: To investigate the effects of Tripteryginm hypoglaucum hutch (THH) on the HPRT gene of human promyelocytic leukemia cells. Methods: HL 60 cells were infected with TTH with different concentrations and put into single cell wells at different time phases. Then the numbers of positive wells were counted and the cell plating efficiency, cloning efficiency and mutation frequency were determined. Results: When the concentration of THH was from 3.35 to 20.10 mg/ml, the cell mutation frequency was positively correlated to the concentration. When the concentration of THH rose up to 33.50 mg/ml, the experiment could not be continued because the cell plating efficiency was too low. This phenomenon might be related with the toxicity of the THH cells. Conclusion: THH can exert mutagenic effects on HPRT gene in HL 60 cells, but there is dose limit in dose response relationship.
