摘要
目的探讨田七注射液对阿霉素(ADR)肾病大鼠转化生长因子β1(TGF-β1)、Smad3蛋白、Smad7蛋白表达的影响。方法采用单侧肾切除加ADR重复注射肾病模型。将健康SPF级SD大鼠随机分为3组:假手术组(n=8)、模型组(n=15)、田七注射液组(n=15)。治疗8周后处死大鼠,观察24h尿蛋白定量、血尿素氮,肌酐及肾脏组织病理改变,同时观察肾组织TGF-β1、Smad3、Smad7的表达。结果田七注射液能减少尿蛋白的排泄,改善肾功能(Bun、Cr),明显降低肾小球硬化指数及肾间质损伤分数,下调TGF-β1、Smad3蛋白的表达,提高Smad7蛋白表达。结论在阿霉素肾病大鼠肾病模型中,田七注射液对肾组织有一定保护作用,其作用机制可能是通过下调TGF-β1、Smad3蛋白的表达,上调Smad7蛋白的表达,干预Smad蛋白介导的细胞内信号转导,从而实现抗肾纤维化。
Objective To explore Tianqi injection on the expression and significance of TGF-β1,Smad3 and Smad7 on adriamycin-induced nephropathy in rats.Methods The model of unilateral nephrectomy and adriamycin-induced nephropathy was used.SD rats were randomly assigned to three groups:sham operation group(n=8),model group(n=15),and Tianqi group(n=15).Three groups were killed after eight weeks.The levels of 24 hours urokinase protein,blood urea nitrogen,creatinine,histological and renal pathological changes were examined,and the expression levels of TGF-β1,Smad3 and Smad7 protein in the nephridial tissue were measured.Results Tianqi injection could statistically decrease urine protein excretion and improve the renal function(Bun,Cr).The glomerular sclerosis index and renal interstitial scores of Tianqi group were significantly decreased.The expressions of TGF-β1,Smad3 protein were statistically significant decreased.The expression of Smad7 was statistically significant increased.Conclusion Tianqi injection significantly prevents against renal injury in the rats with adriamycin-induced nephropathy,it may be intervene in the Smads signal pathway by decreasing the expression of TGF-β1 and Smad3 and increasing the expression of Smad7,thus achieve anti-fibrosis.
